A Practical Glycan Engineering Method For The Preparation Of Homogenous Antibody Conjugates
WARF: P240090US02

Inventors: Weiping Tang, Deqin Cai, Yuan Zhao

The Invention

UW-Madison researchers have developed a simple, two-step process for creating a disaccharide handle useful as a linker in antibody-drug conjugation. The researchers recognized that the Endo-S2 enzyme used to hydrolyze glycan chains from antibodies yields a functional group that can react with an acetal to create the linker. They created an acetal-containing disaccharide via a two-step chemical synthesis using the commercially available LacNAc.

The inventors noted that the linker appears stable in the limited experimentation they have done. They plan to continue to study ADCs they can make with the linker.
